目的:采用星点设计-效应面法优选白藜芦醇固体脂质纳米粒的处方。方法:采用星点设计-效应面优化法筛选处方,以白藜芦醇包封率及载药量作为评价指标,考察脂质用量、表面活性剂用量、药物量对评价的影响。采用热高压均质法制备白藜芦醇固体脂质纳米粒。结果:热高压均质工艺能有效制备固体脂质纳米粒,优选的最佳处方为单硬脂酸甘油酯用量为4.5%、泊洛沙姆用量为6%、白藜芦醇投药量为0.45%,OD值与理论值偏差-4.12%。结论:星点设计-效应面法能有效优选RES-SLN处方。
OBJECTIVE: To optimize the formulation of resveratrol solid lipid nanoparticles by the method of star point design and response surface methodology. Methods: Star point design - response surface optimization method screening prescriptions, resveratrol entrapment efficiency and drug load as an evaluation index, the amount of lipid, the amount of surfactant, the amount of drug on the evaluation. Preparation of resveratrol solid lipid nanoparticles by hot high pressure homogenization. Results: The hot-pressure homogenization process can effectively prepare solid lipid nanoparticles. The best prescription is 4.5% glycerol monostearate, 6% poloxamer and 0.45 resveratrol %, OD value -4.12% deviation from the theoretical value. Conclusion: Star-point design-response surface method can effectively optimize the RES-SLN prescription.
