[目的]研究利用黄瓜作为生物反应器时人胰岛素的表达情况。[方法]通过农杆菌介导法将修饰过的人胰岛素基因(hINS)转入黄瓜,构建表达人胰岛素的生物反应器,再利用PCR和RT-PCR方法检测hINS的表达情况,最后利用NOD小鼠饲喂试验检测表达蛋白的疗效。[结果]研究成功构建了能够稳定表达人胰岛素的生物反应器,目的蛋白表达量占其总可溶性蛋白含量的0.042%~0.190%;NOD小鼠饲喂试验证实,表达产物对NOD小鼠的糖尿病防治具有显著疗效。[结论]该研究表明利用转基因植物作为生物反应器生产医用蛋白和药品具有理论可行性和巨大的利用价值。
[Objective] The research aimed to study the expression of human insulin in cucumber as a bioreactor. [Method] The modified human insulin gene (hINS) was transferred into cucumber by Agrobacterium tumefaciens method to construct a human insulin bioreactor. The expression of hINS was detected by PCR and RT-PCR. Rat feeding test to detect the efficacy of the expressed protein. [Results] The bioreactor capable of stably expressing human insulin was successfully constructed and the target protein was expressed in 0.042% ~ 0.190% of the total soluble protein. The NOD mouse feeding test confirmed that the expression of the target protein in diabetic mice with NOD Prevention has a significant effect. [Conclusion] This study showed that the use of transgenic plants as bioreactors for the production of medical proteins and pharmaceuticals has theoretical feasibility and great value.
