With the further study of the nervous system, in the embryo environment, a large number of neurons produce molecules with attractive and repulsive coordinating functions. Netrins are members of the extracellular protein family that interact with the receptor family to provide a unique dual signal that controls neuronal migration and axonal growth. Netrin-1 is a family-focused molecule that acts not only as a repulsive and attractive bio-signaling molecule. When acting on a specific receptor, a series of events in the cytoplasm will be triggered. In recent years, studies have found that this interaction can participate in the regulation of tumor occurrence and development. This review summarizes the role of Netrin-1 and its receptors in cancer research in recent years, and then explores its mechanism of affecting tumor development.
