目的:探讨体外诱导脐血产生特异性细胞毒淋巴细胞的可行性。方法:通过体外联合细胞因子体外诱导10份脐血单个核细胞分化为树突状细胞(dendritic cells,DCs),同时让DCs细胞负载U937冻融抗原;使成熟DCs刺激同源的脐血T淋巴细胞生成细胞毒性T细胞(cytotoxic T lymphocytes,CTLs)从而进行杀伤效应实验。MTT法测定杀伤活性。结果:10份脐血标本均可培养出形态典型、功能成熟的DCs。经DCs诱导效应细胞DC-CTLs,在不同效靶比对U937细胞系、K562细胞株和对照组中,杀伤率以U937组最高(P<0.05)。结论:特定抗原负载的脐血DCs细胞有特异性的杀伤效应。
Objective: To investigate the feasibility of inducing specific cytotoxic lymphocytes in cord blood in vitro. Methods: Ten cord blood mononuclear cells were induced to differentiate into dendritic cells (DCs) in vitro and in vitro, and DCs were loaded with U937 freeze-thaw antigen. Mature DCs stimulated homologous cord blood T lymphocytes Cells produce cytotoxic T lymphocytes (CTLs) for killing effect experiments. MTT assay for killing activity. Results: All 10 cord blood samples were able to cultivate DCs with typical morphology and function. Conclusion: Specific antigen-loaded cord blood DCs cells have a specific killing effect.
