目的:探讨人血T细胞在异种肾移植(猪→人)间接体内模型中的变化。方法:以模拟人体生理条件下的体外灌注系统,将人新鲜血灌注离体猪肾25只(实验组),人肾12只(对照组);利用流式细胞术,检测灌注前后不同时间内人血T细胞的变化。结果:经猪肾灌注的人血T细胞在灌注一开始即锐减,由灌注前的678±34下降至324±21,下降了约52%;随着灌注时间的延长,其减少的幅度逐渐减弱。而对照组T细胞在灌注前后无明显变化。结论:与同种移植不同,异种器官接通血流一开始,人血T细胞锐减,表明T细胞可能通过与猪肾血管内皮细胞的粘附,参与了异种移植排斥反应。
Objective: To investigate the changes of human blood T cells in an indirect in vivo model of xenotransplantation (pig → human). Methods: To simulate the in vitro perfusion system under human physiological conditions, 25 fresh porcine kidney (experimental group) and 12 human kidney (control group) were perfused with fresh blood. Changes in human blood T cells. Results: The transfused human blood T cells decreased sharply from the beginning of perfusion, from 678 ± 34 before perfusion to 324 ± 21, down about 52%. With the time of perfusion The extent of its decrease gradually weakened. T cells in the control group before and after perfusion no significant change. CONCLUSION: Different from allografts, the blood flow of xenotransplantation starts from the beginning of blood flow, and T cells in human blood drop sharply, indicating that T cells may participate in xenotransplantation rejection through adhesion with porcine renal vascular endothelial cells.
