目的:考察疏肝健脾法肠激安方对腹泻型肠易激综合征(IBS-D)大鼠免疫系统的影响,探讨其调控IBS-D脑-肠轴异常的免疫学机制。方法:取正常SD大鼠采用母子分离、乙酸刺激结合束缚应激的方法制造IBS-D模型,造模成功后将大鼠随机分为模型组,匹维溴铵组(0.018 g·kg~(-1)),疏肝健脾中药肠激安高、中、低剂量组(33.48,16.74,8.37 g·kg~(-1)),每组6只,连续ig 14 d,另取同批次正常大鼠6只作为正常组,同法给予生理盐水。给药结束后观察各组大鼠的日进食量、糖水偏好情况、自主活动3种行为学特征,酶联免疫吸附测定法(ELISA)检测血清中炎症因子白细胞介素-8(IL-8)和IL~(-1)0的含量,实时荧光定量聚合酶链式反应(Real-time PCR)法检测结肠中肿瘤坏死因子-α(TNF-α)及IL~(-1)βmRNA的表达,并测定胸腺系数和脾系数的变化。结果:与正常组比较,IBS-D模型组大鼠的日均进食量、糖水偏好率及自主活动均减少,血清IL-8含量及结肠中TNF-α,IL~(-1)βmRNA表达量明显升高,血清IL~(-1)0含量明显降低,胸腺系数和脾系数明显增高(P<0.05);与模型组比较,各给药组日均进食量、糖水偏好率及自主活动回升,结肠中TNF-αmRNA水平降低,血清IL~(-1)0含量升高(P<0.05),匹维溴铵组和肠激安高、中剂量组IL-8含量和结肠中IL~(-1)βmRNA水平显著降低(P<0.05),而低剂量无统计学差异,各组胸腺系数显著降低(P<0.05),脾系数呈降低趋势,但无统计学差异。结论:疏肝健脾法能够改善IBS-D大鼠的肝郁脾虚状态,并能够调节炎症因子的表达和一定程度地改善免疫器官功能,这可能是其调控IBS-D脑-肠轴功能异常、治疗IBS-D的作用途径之一。
Objective: To investigate the effect of Shugan Jianpi Decoction (GBWD) on the immune system in rats with diarrhea-predominant irritable bowel syndrome (IBS-D), and to explore its immunological mechanism of regulating IBS-D brain-intestinal axis abnormalities. Methods: IBS-D model was established in normal SD rats by means of separation of mother and children and acetic acid stimulation combined with restraint stress. After successful modeling, the rats were randomly divided into model group, piroxicam group (0.018 g · kg ~ (-1) -1)), and the traditional Chinese medicine of Shuganjianpi group were given high, medium and low doses of gavage (33.48,16.74,8.37 g · kg -1) Six normal rats as a normal group, with the law given saline. At the end of the administration, the daily food intake, the preference of sugar and water and the behavior of three kinds of activities were observed. The levels of interleukin-8 (IL-8) and interleukin-8 (IL-8) in sera were detected by enzyme-linked immunosorbent assay And TNF-α and IL-1β mRNA in the colon were detected by real-time PCR, The thymus coefficient and spleen coefficient were measured. Results: Compared with the normal group, the average daily food intake, the preference of sugar and water and the voluntary activity of the rats in IBS-D group were decreased, the contents of IL-8 and TNF-α and IL-1βmRNA (P <0.05). Compared with the model group, the average daily intake of food, the preference of sugar and water and the recovery of spontaneous activity (P <0.05). The levels of IL-8 in colon and the levels of IL-8 in colon and pilos / 1) β mRNA was significantly decreased (P <0.05), while there was no significant difference at low dose (P <0.05). The spleen coefficient decreased but had no statistical difference. Conclusion: Shuganjianpi method can improve IBS-D rats’ state of liver-spleen deficiency and regulate the expression of inflammatory cytokines and to some extent to improve the function of immune organs, which may be its regulation of IBS-D brain-gut dysfunction , One of the ways to treat IBS-D.
