目的:观察琼脂培养MTT法测定卵巢癌对顺氯氨铂(DDP)体外敏感性与临床疗效的关系。方法:用琼脂培养MTT法对26例卵巢癌患者的癌组织标本行药敏测定,同时观察DDP对卵巢癌治疗的临床疗效。结果:卵巢癌接种活细胞数与光密度(OD值)在一定范围内呈线性相关(r=0.9728,P<0.01)。临床缓解组DDP对卵巢癌细胞的相对抑制率(73±5)明显较临床无效组(36±9)高(P<0.01)。卵巢癌对顺铂体外敏感性与临床疗效相关,体内外阳性符合率是88%(16/18),阴性符合率为100%(8/8),总符合率是92%(2/26)。结论:琼脂培养MTT法能为指导临床化疗选药提供一定依据
Objective: To observe the relationship between the in vitro sensitivity of cisplatin (DDP) and the clinical efficacy of ovarian cancer by MTT assay. Methods: MTT assay was used to detect the susceptibility of cancer tissue samples from 26 patients with ovarian cancer. The clinical efficacy of DDP treatment on ovarian cancer was also observed. Results: There was a linear correlation between the number of viable cells and the optical density (OD) of ovarian cancer in a certain range (r=0.9728, P<0.01). The relative inhibition rate of DDP in ovarian cancer cells in the clinical remission group (73±5) was significantly higher than that in the clinically ineffective group (36±9) (P<0.01). The in vitro sensitivity of ovarian cancer to cisplatin was related to clinical efficacy. The in-vivo positive coincidence rate was 88% (16/18), the negative coincidence rate was 100% (8/8), and the overall compliance rate was 92% (2/26) . Conclusion: MTT assay for agar culture can provide a basis for guiding clinical chemotherapy selection
