目的:制备从尖吻蝮蛇蛇毒中分离的纤溶因子的单克隆抗体,为克隆基因提供特异性的探针。方法:将纤溶因子免疫Balb/C小鼠,取脾细胞与SP2/0骨髓瘤细胞融合,筛选稳定分泌抗体的细胞株。采用血纤维蛋白平板法,观察单克隆抗体能否抑制纤溶因子的纤溶作用。结果:获得2株稳定分泌单克隆抗体的细胞株。Western印迹显示该单克隆抗体能特异地与纤溶因子结合,与其它蛋白无交叉反应,此外,这2株单克隆抗体具有抑制纤溶因子溶解人血纤维蛋白的作用。结论:这2株抗体不仅能特异地与纤溶因子结合,而且具有抑制纤溶的作用,为克隆纤溶因子基因及其功能表达提供了特异性的研究工具。
OBJECTIVE: To prepare a monoclonal antibody to fibrinolytic factor isolated from Agkistrodon acutus Venom, and to provide a probe for cloning genes. Methods: Balb / C mice were immunized with fibrinolytic factor. The spleen cells were fused with SP2 / 0 myeloma cells to screen stable cell lines secreting antibodies. Using fibrin plate method, observe the monoclonal antibody can inhibit the fibrinolytic factor fibrinolysis. Results: Two cell lines stably secreting monoclonal antibodies were obtained. Western blotting showed that the monoclonal antibody specifically binds to fibrinolysis without cross-reaction with other proteins. In addition, these two monoclonal antibodies have the effect of inhibiting fibrinolysis of human fibrin. CONCLUSION: These two antibodies can not only bind fibrinolytic factor specifically but also inhibit the fibrinolysis, providing a specific research tool for cloning fibrinolysis genes and their functional expression.
