根据BACTEC 96 0仪分析动物试验模型主要脏器的结核分枝杆菌生长情况 ,分析结核分枝杆菌MPT6 4和ESAT6DNA疫苗预防结核病的效力。将BALB/C小鼠随机分五组 ,分别用A组生理盐水、B组载体质粒、C组卡介苗、D组重组质粒JW 4 30 3 MPT6 4DNA、E组JW4 30 3 ESAT6DNA免疫 9周后 ,用H3 7RV结核分枝杆菌强毒株攻击小鼠。观察攻击一个月和两个月后的结果。发现C ,D ,E组比B组生长天数总体向后推迟 ,生长时间明显集中 ,生长曲峰强度比B组明显减弱 ,C组肝脏中的感染率比D ,E组有所下降。D组肺脏中感染率下降明显。D ,E组鼠脾淋巴细胞转化率较C组高 ,B明显降低。结论 :与B组比 ,小鼠免疫力的短期预防有所增强 ,其中卡介苗前期效果明显 ;MPT6 4 ,ESAT6是持续效果好。
The efficacy of Mycobacterium tuberculosis MPT6 4 and ESAT6 DNA vaccines against tuberculosis was analyzed based on the BACTEC 96 instrument for the analysis of Mycobacterium tuberculosis growth in major organs of animal models. The BALB / C mice were randomly divided into five groups and each group was immunized with Group A saline, Group B vector, Group C BCG, Group D recombinant plasmid JW 4 30 3 MPT6 4 DNA and Group E JW4 30 3 ESAT6 DNA for 9 weeks. H3 7RV Mycobacterium tuberculosis virulent strain challenged mice. Observe the results for one month and two months after the attack. It was found that the days of growth in group C, D and E were generally postponed compared with those in group B, the growth time was obviously concentrated, the peak of growth curve was significantly weaker than that in group B, and the infection rate in group C was lower than that in group D and E. In group D, the infection rate in lung decreased significantly. The conversion rate of splenic lymphocytes in group D and E was higher than that in group C, and B was significantly decreased. CONCLUSION: Compared with group B, short-term immunity of mice is enhanced, and the effect of BCG is obvious. The effect of MPT6 4 and ESAT6 is good.
