目的:研究整合蛋白β1亚基过表达对肝癌细胞SMMC-7721增殖的影响及机制。方法:采用MTT及细胞流式测定观察整合蛋白β1亚基过表达对肝癌细胞SMMC-7721增殖及细胞周期的影响。蛋白质印迹及RT-PCR检测细胞中p27的蛋白及mRNA的表达,并构建p27的RNA干扰质粒,转染整合蛋白β1亚基过表达细胞,再观察p27的表达变化对细胞增殖的影响。结果:整合蛋白β1亚基过表达可以抑制肝癌细胞SMMC-7721的增殖,并将细胞阻滞在细胞周期的G1期。整合蛋白β1亚基过表达可以在蛋白水平上调p27的表达,但并不影响p27的mRNA水平。p27的RNA干扰质粒可明显抑制p27的表达,而且p27表达的下调可以阻止整合蛋白β1亚基过表达所引起的细胞增殖抑制。结论:整合蛋白β1亚基过表达通过上调p27抑制肝癌细胞SMMC-7721的增殖。
Objective: To study the effect of overexpression of integrin β1 subunit on the proliferation of hepatocellular carcinoma cell line SMMC-7721 and its mechanism. Methods: The effects of overexpression of integrin β1 subunit on proliferation and cell cycle of hepatocellular carcinoma cell line SMMC-7721 were observed by MTT assay and flow cytometry. The expression of p27 protein and mRNA was detected by Western blotting and RT-PCR. The p27 RNAi plasmid was constructed and transfected into integrin β1 subunit overexpression cells. The effect of p27 expression on cell proliferation was also observed. Results: The overexpression of integrin β1 subunit could inhibit the proliferation of SMMC-7721 hepatocellular carcinoma cells and block the cells in the G1 phase of the cell cycle. Overexpression of integrin β1 subunit upregulates the expression of p27 at the protein level but does not affect the mRNA level of p27. p27 RNAi plasmid can significantly inhibit the expression of p27, and p27 expression downregulation of integrin β1 subunit can prevent cell proliferation inhibition caused by over-expression. Conclusion: overexpression of integrin β1 inhibits the proliferation of hepatoma SMMC-7721 cells by up-regulating p27.
