目的研究抑癌基因RASSF1A在肝癌组织以及肝癌细胞株中的表达情况及其与临床相关因素间的关系。方法收集24例手术切除的肝癌及其癌旁组织以及4株肝癌细胞株(SMMC7721、HepG2、BEL7402和BEL7703(由国家人类基因组南方中心提供)。用RT-PCR和Northern Blot的方法来分析基因RASSF1A在肝癌组织和肝癌细胞株中的表达情况。结果67%的原发性肝癌组织中未检测到RASSF1A的mRNA,而相应的癌旁组织中仅33%的样本未表达RASSF1A;75%(3/4)肝癌细胞株也未表达RASSF1A。结论RASSF1A在肝癌中是一潜在的抑癌基因,RASSF1A基因的失表达在原发性肝癌的发生发展中起重要作用,检测RASSF1A的表达情况可用于对肝癌的早期发现和早期诊断,结合临床相关因素对肝癌预后评估也有帮助。
Objective To study the expression of tumor suppressor gene RASSF1A in hepatocellular carcinoma (HCC) and hepatocellular carcinoma (HCC) and its relationship with clinically relevant factors. METHODS: Twenty-four resected HCCs and their peritumoral tissues and four hepatocellular carcinoma cell lines (SMMC7721, HepG2, BEL7402 and BEL7703) were obtained from the National Human Genome Southern Center. RT-PCR and Northern Blot were used to analyze the expression of RASSF1A In HCC tissue and hepatocellular carcinoma cell line.Results No mRNA of RASSF1A was detected in 67% of primary HCC tissues, but only 33% of the corresponding paracancerous tissues did not express RASSF1A in 75% (3 / 4) HCC cell line did not express RASSF1A.Conclusion RASSF1A is a potential tumor suppressor gene in hepatocellular carcinoma, and loss of expression of RASSF1A gene plays an important role in the occurrence and development of primary hepatocellular carcinoma.Detecting the expression of RASSF1A can be used to detect hepatocellular carcinoma The early detection and early diagnosis, combined with clinically relevant factors for the prognosis of liver cancer is also helpful.
