探讨银杏叶提取物(extract of Ginkgo biloba,EGB)对于双酚A(Bisphenol A,BPA)活化RBL-2H3(简称:RBL)表达IL-4和TNF-α的影响。利用BPA激活RBL,通过测定β-氨基已糖苷酶和RT-PCR,研究EGB对于活化RBL脱颗粒以及表达IL-4和TNF-α的影响。结果显示,经BPA活化的RBL,加入EGB(浓度为160μg/ml)后,其β-氨基已糖苷酶释放仅为对照的45%,其IL-4mRNA表达量降低到对照的33.5%,TNF-αmRNA表达量降低到对照的26.8%。结果说明EGB极大地抑制了活化RBL的脱颗粒,同时对于活化RBL表达IL-4和TNF-α也具有极大的抑制作用,提示EGB治疗哮喘的作用与其抑制肥大细胞表达IL-4和TNF-α有关。
To investigate the effect of extract of Ginkgo biloba (EGB) on the expression of IL-4 and TNF-α in RBL-2H3 (RBL) activated by bisphenol A (BPA). Using BPA to activate RBL, the effects of EGB on the de-granulation of activated RBL and the expression of IL-4 and TNF-α were investigated by measuring β-aminohexosaminidase and RT-PCR. The results showed that after BPA-activated RBL was added into EGB (concentration of 160μg / ml), the release of β-aminohexosaminidase was only 45% of the control, the expression of IL-4mRNA decreased to 33.5% α mRNA expression decreased to 26.8% of the control. The results showed that EGB greatly inhibited the degranulation of activated RBL, and also had a great inhibitory effect on the expression of IL-4 and TNF-α in activated RBL, suggesting that EGB treatment of asthma and its inhibition of mast cells expressing IL-4 and TNF- α related.
