目的:观察黄芪炙甘草汤对气血两虚型心率失常小鼠血清炎症因子及心肌酶的影响.方法:选取80只心电图正常的小鼠,按体质量随机分为5组,每组16只,即空白对照组、乌头碱组、气血两虚-乌头碱组、黄芪炙甘草汤低剂量组、黄芪炙甘草汤高剂量组.通过眼眶后静脉慢性放血、饥饿、疲劳的综合法,复制成气血两虚的小鼠模型,再通过左侧颈外静脉注射乌头碱,进而制成气血两虚型心律失常小鼠模型.灌胃给予不同剂量的黄芪炙甘草汤进行干预,观察黄芪炙甘草汤高剂量组、低剂量组对小鼠治疗前后超敏C反应蛋白(hypersensitive C-reactive protein,hs-CRP)、肿瘤坏死因子-α(tumornecrosis factor-α,TNF-α)、白细胞介素-2(interleukin-2,IL-2)、IL-6等4个血清炎症因子,心电图及血清心肌酶的影响.结果:各组小鼠一般体征比较:①气血两虚坞头碱组小鼠表现出倦怠、毛枯蓬松、倦卧少动、拱背消瘦、睑结膜苍白、唇色淡白、体质量减轻、食欲下降,而黄芪炙甘草汤高剂量组和低剂量组上述症状都有显著改善;②黄芪炙甘汤能明显升高模型小鼠的hs-CRP、TNF-α、IL-6以及IL-2等血清炎症水平,与空白组和乌头碱组相比,差异有统计学意义(P<0.05);③各组小鼠心肌酶肌酸激酶(ereatine kinase,CK)和乳酸脱氢酶(lactic dehydrogenase,LDH)水平比较:芪炙甘草汤高剂量和低剂量组均能明显降低气血两虚型心律失常小鼠心肌酶CK和LDH的水平,与气血两虚-乌头碱组比较,具有明显的差异性(P<0.05).结论:黄芪炙甘草汤能较好地保护气血两虚型心律失常模型小鼠心脏功能,这可能与提高动物模型的血清炎症因子水平、改善心电图、降低心肌酶的含量等生化指标有关.“,”Objective:To observe the effect of Huangqi Zhigancao Decoction on serum inflammatory cytokines and myocardial enzymes in mice with Qi and Blood deficiency TCM Syndrome.Methods:80 mice with normal ECG were randomly divided into 5 groups according to their body weight,i.e.the control group,aconitine group,Qi and Blood deficiency + aconitine group,low dose group of Astragalus and Zhigancao Decoction group and high dose group of Astragalus and Zhigancao Decoction.Through the integrated method of posterior orbital venous bloodletting,being hunger and being fatigue,the Qi and Blood deficiency mouse modeling was established.Through the left lateral jugular vein injection of aconitine,the Arrhythmia Mellitus mouse model with Qi and Blood deficiency TCM Syndrome was made.Oral administration of different doses of Astragalus and Zhigancao Decoction was made.(HsCRP) and tumor necrosis factor-α (TNF-α),the level of hypersensitive cucumber,interleukin-2 (IL-2),interleukin-6 (IL-6) in mice of the low dose group and the high dose group before and after treatment were observed and measured.Results:The comparisons of the general signs of mice in each group were as follows:①Rats in Qi and Blood deficiency + aconitine group were tired and thin with fluffy hair,less moving,arching back,pale palpebral conjunctiva,pale lips,weight loss and loss of appetite.The symptoms of mice in Huangqi Zhigancao Decoction high and low dose group were significantly improved.②Huangqi Sunbang Decoction could significantly increase the expression of hs-CRP,TNF-α and IL-6 of the model mice.Compared with those of the blank group and the aconitine group,the differences were statistically significant (P < 0.05).③The levels of CK and LDH in Arrhythmia Mellitus Mice with Qi and Blood Deficiency TCM Syndrom were significantly lower than those in the Qi and Blood deficiency TCM Syndrome group (P < 0.05).Conclusion:Astragalus and Radix Glycyrrhizae Decoction can protect the cardiac function of mice with arrhythmia.This may be related to the improving the level of serum inflammatory factors,the improving the electrocardiogram and reducing the content of myocardial enzymes.
