利用高效液相色谱法,分别检测并对比红花药材中羟基红花黄色素A和金橙Ⅱ的浓度,从而建立对比筛选红花样品中金橙Ⅱ的方法。结果在46个红花样品中,推测有26个可疑阳性样品;而通过对比筛选法能将可疑阳性样品数减至2个。经HPLC-MS法确证了对比筛选法的结果。检测羟基红花黄色素A(HSYA)的色谱条件为:甲醇-乙腈-0.01%磷酸溶液(体积比为26∶2∶72)为流动相。检测金橙Ⅱ的色谱条件为:乙腈-0.025 mol/L溶液(40∶60)为流动相。此法相比于常规只检测红花中金橙Ⅱ的方法,筛选效率与精确度更高,并且更经济、适应性更广。
Using high performance liquid chromatography to detect and compare the concentrations of safflower yellow A and golden orange II in safflower, respectively, to establish a method for screening golden safflower Ⅱ in safflower sample. Results Of 46 safflower samples, 26 suspicious positive samples were presumed, while the number of suspicious positive samples could be reduced to 2 by contrast screening. The results of the comparative screening method were confirmed by HPLC-MS. Detection of hydroxysafflor yellow A (HSYA) chromatographic conditions: methanol - acetonitrile - 0.01% phosphoric acid solution (volume ratio of 26: 2: 72) as the mobile phase. The chromatographic conditions for the detection of golden orange II were as follows: acetonitrile-0.025 mol / L solution (40:60) as the mobile phase. Compared with the conventional method of detecting only golden orange in safflower, the method has higher screening efficiency and accuracy, is more economical and has more adaptability.
