目的:探讨全反式视黄醛(at-Ral)对体外培养人子宫内膜异位症(EMT)在位内膜基质细胞迁移和视黄醇脱氢酶(RDH)10mRNA表达的影响。方法:体外分离培养EMT患者在位子宫内膜基质细胞(EMT-ESC)及正常人子宫内膜基质细胞(N-ESC),采用细胞划痕运动分析及real time PCR方法分别观察不同浓度at-Ral对细胞迁移及RDH10mRNA表达情况。结果:N-ESC的迁移距离及RDH10的表达随at-Ral浓度的升高而减少,1,10μmol/L组与对照组比较差异有统计学意义(P<0.05),而EMT-ESC的迁移距离及RDH10的表达随at-Ral浓度的升高而增加,1,10μmol/L组与对照组比较差异有统计学意义(P<0.05)。结论:at-Ral对内异症患者在位子宫内膜基质细胞的迁移及RDH 10mRNA表达有促进作用。
AIM: To investigate the effect of at-Ral on the migration of eutopic endometrial stromal cells and the expression of retinol dehydrogenase (RDH) 10mRNA in human endometriosis (EMT) in vitro. Methods: EMT-ESC and normal human endometrial stromal cells (N-ESC) were isolated and cultured in vitro. Scratch movement assay and real time PCR method were used to observe the effects of different concentrations of at- Ral on cell migration and RDH10mRNA expression. Results: The migration distance and the expression of RDH10 in N-ESC decreased with the increase of the concentration of at-Ral. There was significant difference between 1 and 10μmol / L groups and the control group (P <0.05), while the migration of EMT-ESC The distance and RDH10 expression increased with the increase of the concentration of at-Ral. There was significant difference between 1 and 10μmol / L group and the control group (P <0.05). Conclusion: At-Ral can promote the migration of endometrial stromal cells and the expression of RDH 10mRNA in endometriosis patients.
