目的研究云南元宝枫黄酮对个旧肺鳞癌细胞(YTMLC)凋亡诱导作用及相关基因表达的影响,为开发应用元宝枫黄酮提供实验依据。方法采用四甲基偶氮唑蓝(MTT)比色法检测元宝枫黄酮对YTMLC细胞的生长抑制作用,流式细胞术TUNEL法检测元宝枫黄酮对YTMLC细胞凋亡的影响,RT-PCR检测元宝枫黄酮对YTMLC细胞p53、p21、bcl-2、bax和caspase-3基因表达的影响。结果元宝枫黄酮作用YTMLC细胞48h后,IC50为(108.53±8.22)mg/L。流式细胞术显示元宝枫黄酮既可以诱导YTMLC细胞凋亡,也可以引起YTMLC细胞坏死。元宝枫黄酮对p53、p21、bax和caspase-3的基因表达均有明显上调作用;对bcl-2基因表达有下调作用。结论云南元宝枫黄酮可以抑制个旧肺鳞癌细胞(YTMLC)体外生长,引起肿瘤细胞坏死,诱导肿瘤细胞凋亡,其诱导肿瘤细胞凋亡的可能机制是通过上调p53、p21、bax和caspase-3基因的表达,下调bcl-2基因表达。
Objective To investigate the effects of Yunnan Fengbao flavonoids on the induction of apoptosis and related gene expression in Gejiu lung squamous carcinoma cells (YTMLC), and to provide experimental basis for the development and application of the compounds. Methods MTT colorimetric assay was used to detect the growth inhibition of YTMLC cells by using flavonoids from Acer trichosanthin. The apoptosis of YTMLC cells was detected by TUNEL method and the apoptosis of YTMLC cells was detected by RT-PCR. Effect of Maple flavone on p53, p21, bcl-2, bax and caspase-3 gene expression in YTMLC cells. Results After the cells were treated with the components of Aminophor flavonoids for 48h, the IC50 was (108.53 ± 8.22) mg / L. Flow cytometry showed that A. campestris both induced apoptosis of YTMLC cells and also caused necrosis of YTMLC cells. Alopecoru fruit flavonoids on p53, p21, bax and caspase-3 gene expression were significantly up-regulated; bcl-2 gene expression down-regulation. CONCLUSIONS: Yunnan Fengbao flavonoids can inhibit the growth of YTMLC cells in vitro, induce tumor cell necrosis and induce tumor cell apoptosis. The possible mechanism of inducing tumor cell apoptosis is through up-regulating the expression of p53, p21, bax and caspase-3 Gene expression, down-regulate bcl-2 gene expression.
