目的:观察橙盖鹅膏菌中过氧麦角甾醇对人卵巢癌SKOV3细胞增殖的影响及其可能机制。方法:MTT法测试过氧麦角甾醇对细胞增殖的抑制情况,倒置显微镜下观察细胞形态,DAPI染色法观察细胞核形态,Annexin V/PI双染流式细胞术测定细胞早期凋亡率,PI单染流式细胞术测定细胞周期。结果:在93.95~3003.00μg/ml范围内,不同浓度过氧麦角甾醇对SKOV3细胞增殖具有明显抑制作用。187.90、375.38、750.75μg/ml过氧麦角甾醇作用48小时后,细胞出现边缘皱缩,呈梭形生长,脱落及坏死细胞增多,细胞S期比例、细胞早期凋亡率显著增加。结论:过氧麦角甾醇187.90~750.75μg/ml对人卵巢癌SKOV3细胞增殖具有明显抑制作用,可能与其阻滞细胞周期和诱导细胞凋亡有关。
Objective: To investigate the effect of ergosterol in peroxidation on the proliferation of human ovarian cancer cell line SKOV3 and its possible mechanism. Methods MTT method was used to test the inhibitory effect of ergosterol on cell proliferation. Morphology of cells was observed under inverted microscope. Morphological changes of nuclei were observed by DAPI staining. Cell apoptosis rate was detected by flow cytometry with Annexin V / PI double staining. Flow Cytometry to Determine Cell Cycle. Results: In the range of 93.95 ~ 3003.00 μg / ml, different concentrations of ergosterol had a significant inhibitory effect on SKOV3 cell proliferation. At 48 hours after treated by 187.90, 375.38 and 750.75μg / ml ergosterol, the cells appeared fringed, spindle-shaped growth, exfoliated and necrotic cells increased, cell S phase proportion and cell early apoptosis rate increased significantly. CONCLUSION: 187.90 ~ 750.75μg / ml of ergosterol can significantly inhibit the proliferation of human ovarian cancer cell line SKOV3, which may be related to arresting cell cycle and inducing cell apoptosis.
