目的:探讨罗勒多糖对荷NuTu-19卵巢癌大鼠模型的作用。方法:建立荷NuTu-19卵巢癌大鼠模型,体内观察罗勒多糖对荷瘤大鼠肿瘤生长及转移行为的影响,取大网膜瘤组织,提取总RNA,经逆转录、荧光标记后与R 20 s大鼠表达谱芯片杂交,经Gene P ix Pro 3.0图像处理软件分析。同时取肿瘤组织,进行常规病理学检测。结果:罗勒多糖处理组肿瘤的数量明显少于对照组,肿瘤体积小于、腹水量显著少于对照组(P<0.05);腹腔脏器转移程度积分与对照组有显著差异(P<0.01);同时采用表达谱芯片技术从药物组肿瘤组织中比较筛选出了268条差异表达基因。结论:罗勒多糖可能是一种抗卵巢癌转移的新型中草药,它对卵巢癌显现的独特效果可能是多个药效靶点综合作用的结果,本研究也显示了表达谱芯片技术在药物效应靶点研究中的优势和价值。
Objective: To investigate the effect of basil polysaccharide on NuSTu-19 ovarian cancer rat model. Methods: To establish the model of NuTu-19 ovarian cancer in nude mice. The effects of basil polysaccharide on the tumor growth and metastasis in tumor-bearing rats were observed in vivo. The total RNA was extracted from the omentum and the RNA was reverse transcribed and labeled with R 20 s rat cDNA microarray was analyzed by Gene P ix Pro 3.0 image processing software. At the same time take the tumor tissue, routine pathological examination. Results: The number of tumors in basilar polysaccharide treatment group was significantly less than that in control group. The volume of tumor was less than that of control group, and the amount of ascites was significantly lower than that of control group (P <0.05). The integral of metastasis of abdominal organ was significantly different from that of control group (P <0.01). At the same time, 268 differentially expressed genes were screened out from the tumor tissue of the drug using expression profiling chip technology. Conclusion: Basil polysaccharides may be a new type of Chinese herbal medicine against ovarian cancer metastasis. The unique effect of basil polysaccharide on ovarian cancer may be the result of the combination of multiple pharmacodynamic targets. This study also shows that the expression profiling of microarray in drug target Point in the study of the advantages and values.
