目的探讨生肌玉红胶原促进慢性缺血性创面愈合的可能作用机制。方法 72只大鼠随机分为空白组、对照组、实验组3组,每组24只。所有大鼠建立大鼠后肢缺血性创面模型,空白组直接缝合皮肤,实验组以生肌玉红提取液(0.5 g/ml,50μl/只)载入10 mm×10 mm×2 mm长方体胶原放置血管离断缺损处的缺血肌肉组织处,对照组将载入50μl生理盐水的相同大小胶原放置于血管离断缺损处的缺血肌肉组织处,然后缝合皮肤。各组均观察28天。观察各组大鼠一般情况,并分别于第3、7、14、28天检测大鼠后肢缺血创面处微血管计数及对照组与实验组植入标本内血红蛋白含量。结果空白组大鼠后肢均有跛行,部分苍白并见到肢端青紫,实验组大鼠后肢无明显缺血表现。与空白组同时间点比较,对照组第14、28天时微血管计数高于空白组,实验组第7、14、28天时亦明显高于空白组(P<0.05)。与对照组同时间点比较,实验组第7、14天时微血管计数明显升高(P<0.05)。实验组第3、7、14、28天植入标本内血红蛋白含量明显高于同时间点对照组(P<0.05)。结论生肌玉红胶原可促进缺血性创面组织血管新生并改善其血流灌注,可能为其促进慢性缺血性创面愈合的作用机制。
Objective To investigate the possible mechanism of myogenic jade red collagen in promoting chronic ischemic wound healing. Methods 72 rats were randomly divided into blank group, control group and experimental group, with 24 rats in each group. The model of hindlimb ischemic wound was established in all the rats, and the blank group was used to suture the skin directly. In the experimental group, 10g × 10mm × 2mm collagen was loaded into the muscle of Yujian jade red extract (0.5g / ml, 50μl / body) The ischemic muscle tissue at the vascular defect was placed and the control group placed the same size collagen loaded with 50 μl of physiological saline on the ischemic muscle tissue at the vascular defect and the skin was sutured. All groups were observed for 28 days. The general condition of rats in each group was observed. The counts of microvessels in ischemic hind limbs and hemoglobin in control group and experimental group were detected on the 3rd, 7th, 14th and 28th day respectively. Results In the blank group, the hindlimbs were all limp, partially pale and bruising. The hindlimbs in the experimental group showed no obvious ischemia. Compared with the blank group, the microvessel counts of the 14th and 28th day in the control group were significantly higher than those in the blank group. The experimental group was also significantly higher than the blank group on the 7th, 14th and 28th days (P <0.05). Compared with the control group at the same time points, the experimental group at the 7th and 14th day, the microvessel count was significantly higher (P <0.05). The hemoglobin content in the experimental group at 3, 7, 14 and 28 days was significantly higher than that in the control group at the same time point (P <0.05). Conclusion Myogenic jade red collagen can promote angiogenesis and improve perfusion of ischemic wound tissue, which may be the mechanism of its action in promoting chronic ischemic wound healing.
