目的观察青白散对慢性软组织损伤大鼠骨骼肌中Ⅰ、Ⅲ型胶原蛋白表达的影响以及局部软组织的形态学改变。方法 54只雄性SD大鼠随机分为自然愈合组、云南白药组、青白散组,每组18只。采用慢性软组织损伤动物模型,分别予以生理盐水(10ml/kg)、云南白药(0.18g/kg)和青白散(0.54g/kg)灌胃,每日1次,连续14d。给药后第1周末、第2周末、第3周末,免疫组织化学方法检测大鼠慢性软组织损伤后骨骼肌中ⅠⅢ型胶原蛋白的表达,酶联免疫法检测骨骼肌中肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)含量的动态变化。结果青白散组肌肉再生速度、愈合质量显著优于自然愈合组,在不同时间点的骨骼肌中TNF-α和IL-6的含量均较自然愈合组显著降低(P<0.01),Ⅰ、Ⅲ型胶原蛋白的表达均较自然愈合组显著降低(P<0.01)。结论青白散能阻止胶原纤维的过度生成,降低致炎因子水平,从而有利于损伤组织的修复。
Objective To observe the effect of Qingbai San on the expression of collagen type Ⅰ and Ⅲ in skeletal muscle of rats with chronic soft tissue injury and the morphological changes of local soft tissue. Methods Fifty-four male SD rats were randomly divided into natural healing group, Yunnan Baiyao group and Qingbai powder group, with 18 rats in each group. The animal model of chronic soft tissue injury was used. The rats were gavaged with normal saline (10ml / kg), Yunnanbaiyao (0.18g / kg) and Qingbai San (0.54g / kg) once daily for 14 days. At the first weekend, the second weekend and the third weekend after the administration, the expression of type I collagen in skeletal muscle of rats with chronic soft tissue injury was detected by immunohistochemical method. The levels of tumor necrosis factor-alpha TNF-α) and interleukin-6 (IL-6) levels in rats. Results The muscle regeneration rate and healing quality of Qingbai powder group were significantly better than those of natural healing group. The levels of TNF-α and IL-6 in skeletal muscle of rats in Qingbai San group were significantly lower than those in natural healing group (P <0.01) Collagen expression was significantly lower than the natural healing group (P <0.01). Conclusion Qingbai San can prevent the excessive formation of collagen fibers, reduce the level of pro-inflammatory cytokines, which is conducive to the repair of damaged tissue.
