在粮食、蔬菜等食品中,由于人类和动物的排泄物或在运输过程中的污染而含有尿素。常用的测定方法是微量扩散法,但此法在基层单位常因缺少脲酶而使实验遇到困难。本法参考了美国出版的《分析毒理学方法手册》中关于尿素的测定方法。并用新华层析滤纸代替纤维素板。测定效果良好,5微克可见清晰斑点,现介绍如下,供参考。一、原理:用丙酮提取样品中尿素,以纸层析法分离尿素,再与对-二甲氨基苯甲醛(P-Dimethylaminobenzaldchyde)反应,呈现黄绿色斑点,与标准点比较,概略定量
