高效液相色谱法测定脱落酸与3-吲哚乙酸是目前最常用的方法之一,根据文献报导,该法最常用的色谱柱为各种牌号的C_(18)柱,流动相有甲醇-醋酸水溶液,甲醇-乙腈,乙腈-醋酸(含有醋酸铵)的缓冲液,甲醇-磷酸(pH=3)缓冲液,甲醇-磷酸缓冲液(pH=6.5含有0.01mol·1~(-1)TBA),甲醇-K_2HPO_4(含有四丁基铵)等,结合我们的条件,我们做了一些对比试验,认为甲醇—磷酸缓冲溶液(pH=7.2,用0.0025mo1·1~(-1)的K_2HPO_4与KH_2PO_4配制)效果较好。
Determination of abscisic acid and 3-indoleacetic acid by high performance liquid chromatography is one of the most commonly used methods. According to the literature, the most commonly used columns for this method are C 18 columns of various grades with a mobile phase of methanol- Aqueous solutions of acetic acid, methanol-acetonitrile, acetonitrile-acetic acid (containing ammonium acetate), methanol-phosphate buffer (pH = 3), methanol-phosphate buffer ), Methanol-K_2HPO_4 (containing tetrabutylammonium) and so on. According to our conditions, we made some comparative experiments. It is concluded that methanol-phosphoric acid buffer solution (pH 7.2, K_2HPO_4 with 0.0025mo_1-1 KH_2PO_4 preparation) is better.
