从非胰岛素依赖型糖尿病(NIDDM)患者外周血的白细胞中提取DNA.以胰岛素基因5’-末端上游的某一DNA序列为引物,对特定的靶序列进行体外扩增,即TaqDNA聚合酶链反应(PCR),将其扩增产物进行电泳分离,在紫外分析仪下观察结果并与正常人对照,发现16例NIDDM的M患者胰岛素基因有所改变,提示此型糖尿病患者的胰岛素基因改变可能是导致NIDDM遗传因素之一。
DNA was extracted from leukocytes in the peripheral blood of non-insulin dependent diabetes mellitus (NIDDM) patients. A certain DNA sequence upstream of the 5’-end of the insulin gene was used as a primer to amplify a specific target sequence in vitro, that is, Taq DNA polymerase chain reaction (PCR), and the amplified product was electrophoresed and separated under an ultraviolet analyzer The results of the observation and comparison with normal people found that 16 cases of M patients with NIDDM insulin gene has changed, suggesting that insulin resistance in this type of diabetic patients may be one of the genetic factors leading to NIDDM.
