严重急性呼吸综合征(SARS)疫情在北京造成了严重后果,冠状病毒已被广泛认定为导致SARS的病原体。但是临床和实验提示,SARS病因可能不是单一冠状病毒感染。本文报道了我们从北京市第1例SARS患者和其母亲的咽拭子中分离出呼肠病毒。在电子显微镜下,可观察到典型的呼肠病毒。血清学实验显示,38例SARS患者中24例对呼肠病毒呈阳性反应。针对呼肠病毒的基因保守序列(S2基因片段)设计引物,RT-PCR实验扩增出特异性DNA片段。进一步DNA序列分析表明是一种独特的呼肠病毒(呼肠病毒科正呼肠病毒属)。初步动物实验显示,该呼肠病毒可导致非典型样肺炎发生和小鼠死亡,尽管如此,呼肠病毒感染是否与SARS疫情有关,还有待进一步研究才能阐明。
Severe acute respiratory syndrome (SARS) outbreaks have had serious consequences in Beijing, and the coronavirus has been widely recognized as the causative agent of SARS. However, clinical and experimental evidence suggests that the cause of SARS may not be a single coronavirus infection. This article reports that we isolated the reovirus from the throat swab of the first SARS patient in Beijing and his mother. Under the electron microscope, a typical reovirus can be observed. Serological tests showed that 24 of 38 SARS patients were positive for reovirus. Primers were designed based on the gene conserved sequence of reovirus (S2 gene fragment), and specific DNA fragments were amplified by RT-PCR. Further DNA sequence analysis revealed a unique reovirus (Reovirus family). Preliminary animal experiments show that the reovirus can cause the death of atypical pneumonia and mice. However, whether reovirus is associated with SARS remains to be elucidated.
