目的研究动脉粥样硬化大鼠动脉组织中丝氨酸(Src)抑制的C激酶底物(SSeCKS)的表达。方法以高脂饲料喂养加维生素D3腹腔注射建立SD大鼠动脉粥样硬化模型(A),对照(B)组,每组8只给予基础饲料喂养。测定血清总胆固醇(TC)及甘油三酯(TG);病理学观察HE染色的主动脉;用免疫组织化学法及免疫印迹法检测SSeCKS在动脉中的表达,免疫荧光双标法观察SSeCKS的细胞定位。结果12周时A组TC和TG较B组明显升高(P<0.05);A组形成纤维增生性动脉粥样硬化病变,SSeCKS表达显著增强,SSeCKS表达主要分布在动脉内膜、中膜,部分定位于内皮细胞。结论高脂饮食所致动脉粥样硬化大鼠主动脉SSeCKS表达增加,SSeCKS可能参与了血管重塑及细胞凋亡从而促进动脉粥样硬化的发展。
Objective To investigate the expression of serine (Src) -dependent C-kinase substrate (SSeCKS) in atherosclerosis rats. Methods The model of atherosclerosis in SD rats was established by intraperitoneal injection of high fat diet and vitamin D3 (A), and the control group (B), 8 rats in each group were fed with basal diet. Serum total cholesterol (TC) and triglyceride (TG) were measured. The aorta of HE staining was observed by pathology. The expression of SSeCKS in artery was detected by immunohistochemistry and Western blotting. Positioning. Results At 12 weeks, TC and TG in group A were significantly higher than those in group B (P <0.05). Group A showed fibrotic atherosclerotic lesions and SSeCKS expression was significantly increased. SSeCKS expression mainly distributed in arterial intima, tunica media, Part of the localization of endothelial cells. Conclusions The expression of SSeCKS in the aorta of atherosclerosis rats induced by high fat diet is increased. SSeCKS may be involved in vascular remodeling and apoptosis so as to promote the development of atherosclerosis.
