目的:建立通痹胶囊乙酸乙酯部位的HPLC色谱指纹图谱测定方法。方法:采用HPLC法,Thermo Syncronis C18色谱柱(250mm×4.6mm,5μm)、检测波长290nm、流速0.8m L/min、柱温30℃、进样量10μL进行梯度洗脱,对13批通痹胶囊的乙酸乙酯部位样品进行指纹图谱测定。结果:确定葛根素为内标物,建立了生物碱类成分的HPLC对照指纹图谱,标定了21个共有峰,确定了2个共有峰,基本确定了17个色谱峰的药材归属,占总共有峰数的80.95%。结论:该方法建立的指纹图谱灵敏度高,稳定性好,可为通痹胶囊的质量控制提供可靠依据。
Objective: To establish a method for the determination of ethyl acetate in Tongbai capsule by HPLC chromatographic fingerprinting. Methods: The HPLC method was used on a Thermo Syncronis C18 column (250 mm × 4.6 mm, 5 μm) with a detection wavelength of 290 nm and a flow rate of 0.8 m L / min. The column temperature was 30 ℃ and the injection volume was 10 μL. The ethyl acetate fraction of the capsule was subjected to fingerprinting. Results: The HPLC fingerprint of alkaloids was established by using puerarin as internal standard. The common peaks of 21 alkaloids were calibrated and two common peaks were identified, and the identification of 17 medicinal materials was basically confirmed 80.95% of the peaks Conclusion: The fingerprinting established by this method has high sensitivity and good stability, which can provide a reliable basis for the quality control of Tongbai capsule.
