目的:建立弗朗鼠李皮的指纹图谱及含量测定方法。方法:指纹图谱采用HPLC法,Phenomenex luna C18(250 mm×4.6 mm,5μm)色谱柱;流动相为甲醇(A)-0.1%磷酸溶液(B),梯度洗脱;流速1.0 mL/min;柱温30℃;检测波长264 nm。含量测定采用HPLC法,流动相为甲醇-0.1%磷酸溶液(70∶30);检测波长254 nm。结果:指纹图谱重现性良好;由于没有稳定的对照品,所以对其水解产物大黄素进行含量测定,进样量在0.0383~0.6511 mg范围内呈良好的线性关系(r=1.0000),平均加样回收率为102.20%,RSD为1.3%。结论:该方法简便易行,重复性好,可用于弗朗鼠李皮的质量控制。
OBJECTIVE: To establish a fingerprint and content determination method of Furomilus wallichii skin. METHODS: The fingerprint was characterized by HPLC on a Phenomenex luna C18 (250 mm × 4.6 mm, 5 μm) column with a mobile phase of methanol (A) -0.1% phosphoric acid (B), gradient elution at a flow rate of 1.0 mL / Temperature 30 ℃; detection wavelength 264 nm. Determination of content by HPLC method, the mobile phase methanol -0.1% phosphoric acid solution (70:30); detection wavelength 254 nm. Results: The fingerprint was good reproducibility. Because there was no stable reference substance, the emodin content of the hydrolyzate was determined. The linear range was 0.0383 ~ 0.6511 mg (r = 1.0000) The sample recovery was 102.20% with a RSD of 1.3%. Conclusion: The method is simple, reproducible and can be used for the quality control of Furong walnut skin.
