目的:观察海藻玉壶汤对实验性自身免疫甲状腺炎大鼠的保护作用。方法:45只成年雌性SD大鼠随机分为对照组、模型组和治疗组。采用多点皮下注射猪甲状腺球蛋白(PTg)和高碘饮水喂养进行实验性自身免疫甲状腺炎大鼠造模,对照组注射等量生理盐水。治疗组采用海藻玉壶汤灌胃给药,1次/d,连续6周;对照组和模型组同频率生理盐水灌胃。末次给药后大鼠眼眶取血测定血清甲状腺球蛋白抗体(TGAb)、甲状腺过氧化物酶抗体(TPOAb)、三碘甲状腺原氨酸(T3)和四碘甲状腺原氨酸(T4)水平;光镜下观察甲状腺组织病理形态学的变化情况。结果:与对照组相比,模型组和治疗组TPOAb、T3和T4水平显著升高(P<0.05),模型组Tg Ab较对照组升高,差异有统计学意义(P<0.05)。治疗组Tg Ab、TPOAb、T3和T4水平较模型组显著下降,差异有统计学意义(P<0.05)。治疗组甲状腺组织免疫组化结果显示甲状腺滤泡大小适中、规则,结构较完整,染色颗粒分布减少,可改善甲状腺炎性细胞损伤。结论:海藻玉壶汤可调节EAT模型大鼠血清甲状腺球蛋白抗体、甲状腺过氧化物酶抗体、三碘甲状腺原氨酸和四碘甲状腺原氨酸水平,具有减轻甲状腺病理损伤的作用。
Objective: To observe the protective effect of seaweed Yuhu soup on experimental autoimmune thyroiditis in rats. Methods: Forty-five adult female Sprague-Dawley rats were randomly divided into control group, model group and treatment group. The experimental autoimmune thyroiditis model was induced by multiple subcutaneous injection of pig thyroglobulin (PTg) and high iodine drinking water, and the control group was injected with the same amount of normal saline. In the treatment group, seaweed juguitang was administered intragastrically once a day for 6 weeks, while the control group and the model group were administered with normal saline. Serum thyroglobulin antibody (TGAb), thyroid peroxidase antibody (TPOAb), triiodothyronine (T3) and tetrathionine (T4) levels were measured in the orbital tissues after the last administration. Under light microscope, the histopathological changes of thyroid tissue were observed. Results: Compared with the control group, the levels of TPOAb, T3 and T4 in the model group and the treatment group were significantly increased (P <0.05). The Tg Ab in the model group was significantly higher than that in the control group (P <0.05). The levels of Tg Ab, TPOAb, T3 and T4 in the treatment group were significantly lower than those in the model group (P <0.05). Thyroid tissue immunohistochemistry results showed that thyroid follicular size is moderate, the rules, the structure is more complete, reduce the distribution of stained particles, can improve thyroid inflammatory cell injury. CONCLUSION: Seaweed Yuhu Tang can regulate serum thyroglobulin antibody, thyroid peroxidase antibody, triiodothyronine and tetraiodothyronine in rats with EAT model, which can alleviate the pathological damage of thyroid.
