补肾填精方对去卵巢大鼠炎症因子与破骨细胞活性的影响

目的:对比观察补肾填精方左归丸与左归丸去补阳药对去卵巢骨质疏松模型大鼠炎症因子和破骨细胞活性的影响,探讨左归丸阳中求阴配伍的作用机制。方法:雌性SD大鼠32只,双侧卵巢摘除术建立骨质疏松模型,分为假手术组、模型对照组、左归丸组、左归丸去补阳药组4组,每组8只。灌胃给药90 d,收集各组大鼠血清和股骨组织,采用HE染色法观察股骨组织形态学,采用TRAP染色法检测股骨组织破骨细胞数,采用ELISA法检测血清中TNF-α、IL17和RANKL水平。结果:与模型对照组比较,补肾填精方改善去卵巢大鼠股骨组织形态学,减少破骨细胞数量;显著降低去卵巢大鼠血清TNF-α、IL-17、RANKL水平(P<0.05)。结论:补肾填精方能够抗去卵巢骨质疏松症大鼠骨丢失,其作用机制可能与其通过提高雌激素水平以调节炎症因子抑制破骨细胞活性有关,全方效果较为明显。 OBJECTIVE: To compare the effects of Zuogui Pill and Zuogui Pill on the inflammatory cytokines and osteoclast activity in ovariectomized osteoporosis model rats, and to explore the mechanism of compatibility of Zuogui Pill . METHODS: Thirty-two female Sprague-Dawley rats were divided into 4 groups: sham operation group, model control group, Zuoguiwan group and Zuoguiwan group, 8 in each group . Serum and femur tissues of rats in each group were collected by gavage for 90 days. The morphology of femur was observed by HE staining. The number of osteoclasts in femur was detected by TRAP staining. The levels of TNF-α, IL17 And RANKL levels. Results: Compared with the model control group, Bushen Tianjing Decoction could improve the femoral histomorphology and decrease the number of osteoclasts in ovariectomized rats, and significantly decrease the levels of TNF-α, IL-17 and RANKL in ovariectomized rats (P <0.05) . Conclusion: Bushenfangjing Recipe can prevent bone loss in ovariectomized rats with osteoporosis. Its mechanism may be related to the inhibition of osteoclast activity by regulating the level of estrogen to regulate inflammatory cytokines. The effect of all prescriptions is more obvious.
