目的观察榄香烯对裸鼠胃癌移植瘤的作用并探讨其相关机制。方法雄性裸鼠建立人胃癌SCG-7901细胞裸鼠移植瘤模型,随机分为对照组、榄香烯组、PD98059组、榄香烯和PD98059联合组(榄香烯+PD98059组)。对比榄香烯和PD98059对裸鼠胃癌移植瘤的作用,测量对比裸鼠胃癌移植瘤的大小及药物对裸鼠生长的抑制作用。Western-blot方法检测各组ERK1/2蛋白、p-ERK1/2、p-P38蛋白的表达情况。结果 3个实验组在治疗后移植瘤大小与对照组比较明显减小,榄香烯组较对照组p-ERK1/2蛋白表达显著增多,3个治疗组相比对照组p-p38蛋白表达显著增加,两药联合比单一药物作用显著(均P<0.05或P<0.01)。结论榄香烯和PD98059都能促进肿瘤组织凋亡,其中榄香烯促进裸鼠胃癌移植瘤凋亡可能与p-ERK1/2蛋白和p-p38蛋白上调有关,而PD98059促进裸鼠胃癌移植瘤凋亡可能与p-P38蛋白上调有关。
Objective To observe the effect of elemene on gastric cancer xenografts in nude mice and to explore its related mechanism. Methods The nude mice model of human gastric cancer SCG-7901 cells was established in male nude mice and randomly divided into control group, elemene group, PD98059 group, elemene and PD98059 group (elemene + PD98059 group). The effect of elemene and PD98059 on the transplanted tumor of gastric cancer in nude mice was compared. The size of transplanted tumor of gastric cancer in nude mice was measured and the inhibitory effect of drugs on the growth of nude mice was measured. The expression of ERK1 / 2, p-ERK1 / 2 and p-P38 in each group were detected by Western-blot. Results After treatment, the size of xenografts in the three experimental groups was significantly reduced compared with the control group. The expression of p-ERK1 / 2 in the elemene group was significantly increased compared with the control group. Conclusion Elemene and PD98059 can both promote the apoptosis of tumor tissue.
