生发中心(germinal center,GC)支持B细胞竞争与亲和力成熟的过程。滤泡辅助T细胞通过与B细胞长时程互作来传递辅助信号并促进生发中心形成。然而,当生发中心形成以后,T细胞如何调控B细胞间的竞争及选择尚不明确。借助骨髓嵌合体和活体成像研究发现,GC里T-B互作短暂而重复,由ICOS-ICOSL分子以一种正反馈的方式调控,而这种正反馈对于迅速、有效地筛选出高亲和力抗体至关重要。这些结果使我们更进一步理解了生发中心亲和力选择的机制。
The germinal center (GC) supports the process of B cell competition and affinity maturation. Follicular helper T cells transmit helper signals and promote germinal center formation by interacting with long-term B cells. However, when the germinal center is formed, it is unclear how T cells regulate the competition and selection among B cells. With bone marrow chimerism and in vivo imaging studies, it was found that TB interactions in GC were transient and repetitive and regulated by ICOS-ICOSL molecules in a positive feedback manner. This positive feedback is very important for the rapid and effective screening of high affinity antibodies important. These results give us a better understanding of the mechanism of affinity selection in germinal centers.
