建立了HPLC法测定肠循环灌注液中的炔丙基半胱氨酸,以酚红进行体积校正,采用C18色谱柱,以乙腈-水为流动相,梯度洗脱,双波长(220和362 nm)检测。炔丙基半胱氨酸和酚红在12.5~800μg/ml和0.63~40μg/ml浓度范围内线性关系良好。采用大鼠在体肠循环灌注模型研究炔丙基半胱氨酸的吸收机制,结果显示,不同pH下炔丙基半胱氨酸的吸收速率常数(Ka)和表观渗透系数(Papp)存在显著性差异(P<0.05)。
A HPLC method was developed for the determination of propargylcysteine in intestinal perfusate. The volume was calibrated with phenol red. The mobile phase consisted of acetonitrile and water with gradient elution on a C18 column with dual wavelength (220 and 362 nm ) Test. Propargylcysteine and phenol red had good linearity in the range of 12.5 ~ 800μg / ml and 0.63 ~ 40μg / ml. The rat model of intestinal perfusion was used to study the mechanism of propargylcysteine absorption. The results showed that the propargylcysteine absorption rate constant (Ka) and apparent permeability coefficient (Papp) Significant difference (P <0.05).
