薄层层析是一项重要的分离分析技术。自从Randrath应用ECTEOLA纤维素薄层分离AMP、ADP和AFP等化合物以来,这一技术已广泛应用于核酸类化合物的分离。Josefsson利用荧光物质提高灵敏度的方法对碱基和单核苷酸进行了微量的半定量测定。我们采用DEAE纤维素薄层,以稀HCl为层层剂,较满意地分离了AMP、GMP、CMP和UMP四种核糖单核苷酸,并得到较好的定量回收率。
Thin layer chromatography is an important separation and analysis technique. This technique has been used extensively in the isolation of nucleic acid compounds since Randrath used compounds such as AMP, ADP and AFP to separate ECTEOLA cellulose sheets. Josefsson used a method of increasing the sensitivity of the fluorescent substance to make a small amount of semi-quantitative determination of bases and single nucleotides. We use DEAE cellulose thin layer, dilute HCl layer agent, more satisfactory separation of AMP, GMP, CMP and UMP four kinds of ribose single nucleotide, and get better quantitative recovery.
