目的研究黄芩黄酮化合物bw-lzj-6504、bw-lzj-6517a在体外对肾透明细胞癌细胞增殖的抑制作用。方法磺酰罗丹明B(SRB)比色法测定2种化合物对786-0肾透明细胞癌细胞增殖的影响及半数抑制浓度(IC50);Western blot法检测2种化合物对细胞中多聚(ADP-核糖)聚合酶1(PARP1)、caspase-3、剪切型caspase-3、微管相关蛋白1轻链3(LC3)、P62蛋白表达水平的影响。结果两种黄芩黄酮化合物bw-lzj-6504、bw-lzj-6517a都能显著抑制786-0细胞的增殖、促进其死亡。同时bw-lzj-6504、bw-lzj-6517a能显著降低PARP1、caspase-3蛋白水平,增加PARP1、剪切型caspase-3蛋白水平、对LC3-Ⅱ蛋白水平无明显影响。结论两种黄芩黄酮化合物通过促进细胞凋亡抑制肾透明细胞癌细胞增殖。
Objective To study the inhibitory effect of bw-lzj-6504 and bw-lzj-6517a on the proliferation of renal clear cell carcinoma cells in vitro. Methods The inhibitory concentration (IC50) and the half inhibitory concentration (IC50) of two compounds on the proliferation of 786-0 renal clear cell carcinoma were determined by sulforhodamine B (SRB) colorimetric assay. The effects of two compounds on the proliferation of 786-0 renal clear cell carcinoma - ribose) polymerase 1 (PARP1), caspase - 3, cleaved caspase - 3, microtubule - associated protein 1 light chain 3 (LC3) and P62 protein. Results Both bw-lzj-6504 and bw-lzj-6517a could significantly inhibit the proliferation and promote the death of 786-0 cells. At the same time, bw-lzj-6504 and bw-lzj-6517a could significantly decrease PARP1 and caspase-3 protein levels and increase PARP1 and cleaved caspase-3 protein levels, but had no significant effect on LC3-Ⅱ protein levels. Conclusions Both skullcap flavonoids can inhibit the proliferation of renal clear cell carcinoma cells by promoting apoptosis.
