目的 探讨甲基对硫磷 (M16 0 5 )简便、敏感、特异的免疫分析方法。方法 矩阵法选择好包被抗原和单克隆抗体最佳条件 ,建立M16 0 5间接竞争抑制ELISA检测法 ,并分别检测M16 0 5染毒小鼠和接触工人血清中M16 0 5含量。结果 该方法线性范围为 5~ 5 0 0ng/ml ,最低检出浓度为 5ng/ml ,组内和组间变异系数不大于 11 5 %和 14 6 % ,回收率平均为 96 2 %。M16 0 5染毒小鼠和作业工人血清中M16 0 5检出量及检出率与染毒剂量、检测时间及作业工种有关。结论 初步认为此法可用于M16 0 5接触生物标志物的检测。
Objective To investigate a simple, sensitive and specific immunoassay for methyl parathion (M16 0 5). The optimal conditions of coating antigen and monoclonal antibody were selected by the method of matrix method. The indirect competitive inhibition ELISA (M16 05) assay was established to detect M16 0 5 levels in M16 05 and M16 0 5 exposed workers. Results The linear range of this method was 5 ~ 500 ng / ml. The lowest detection limit was 5 ng / ml. The coefficient of variation (CV) within and between groups was no more than 11 5% and 14 6%. The average recovery was 96 2%. The detection rate and detection rate of M16 0 5 in the serum of M16 0 5-exposed mice and workers were related to the dosage, testing time and working type. Conclusion This method can be used to detect M16 0 5 biomarkers.
