目的 探讨巯基乙醇和维生素C对镉孵育红细胞GSH Px活性的影响。方法 用酶联法分别测定对照组RBC、Cd孵育RBC、巯基乙醇 +Cd孵育RBC、VitC +Cd孵育RBC的谷胱甘肽过氧化物酶 (GSH Px)活性。结果 ①Cd孵育RBC的GSH Px活性显著低于对照组 (P <0 0 1) ;②巯基乙醇 +Cd孵育RBC的GSH Px活性与Cd孵育RBC的GSH Px活性无显著性差异 ;③VitC +Cd孵育RBC的GSH Px活性显著高于Cd孵育组 (P <0 0 1)。结论 ①加入到抗凝血中的Cd易进入RBC中 ,并抑制RBC的GSH Px活性 ,且随进入到RBC中的Cd浓度的增加而增加 ;②巯基乙醇( 2 μl/ml血 )不能有效地阻断Cd对RBC的GSH Px活性的抑制 ;③VitC可显著改善Cd对RBC的GSH Px活性的抑制作用。
Objective To investigate the effects of mercaptoethanol and vitamin C on the activity of GSH Px in cadmium-incubated erythrocytes. Methods The activities of glutathione peroxidase (GSH Px) in RBC, RBC incubated with Cd, RBC incubated with mercaptoethanol and Cd, RBC incubated with Cd and VitC + Cd were determined by enzyme-linked immunosorbent assay. Results ① GSH Px activity of RBC incubated with Cd was significantly lower than that of control (P <0.01); ② GSH Px activity of RBC incubated with mercaptoethanol + Cd was not significantly different from that of GSH Px incubated with Cd; GSH Px activity was significantly higher than that of Cd incubation group (P <0.01). Conclusions ① Cd added into anticoagulant can easily enter RBC and inhibit the GSH Px activity of RBC, and increase with the increase of Cd concentration entering into RBC; ② Mercaptoethanol (2 μl / ml blood) can not effectively Blocking the inhibition of Cd on RBC GSH Px activity; VitC can significantly improve the inhibitory effect of Cd on RBC GSH Px activity.
