通过硫酸铵分级分离,DEAE—交换纤维素柱层析及SephadexG-200凝胶过泸等方法,从厦门文昌鱼中部份提纯一种偏碱性蛋白酶,并对该酶的某些性质作了初步的研究.该酶作用于酪蛋白的最适pH为7.0,最适温度30℃,测得其米代常数(K_M)值为3.08×10~(-4)M,而pH不影响其K_M值。对几种蛋白质的水解速度不同,其中对血红红蛋白的水解速度最快。某些化学试剂如半胱氨酸及谷胱甘肽(都为10~(-3)M)能增进酶活力,而P-CMB及碘代乙酰胺(都为10~(-3)M)则比较明显的降低酶活力。
