顶端外胚层嵴(AER)是中胚层间质细胞诱导其外侧的外胚层细胞形成突起结构,位于肢芽的远端边缘背腹交界处,是肢体生长发育的主要信号中心。利用转基因技术构建在AER中表达Cre重组酶的转基因小鼠(Col10a1-Cre),使Cre重组酶在AER中特异性表达,从而可以特异性地在AER中敲除被LoxP序列锚定的目的基因。确定此转基因小鼠Cre重组酶的表达的时间和空间特异性。实验结果证明Cre重组酶在顶端外胚层嵴中能够特异性表达并能介导LoxP间的重组。在顶端外胚层嵴发现特异性的蓝染,将Cre重组酶的表达范围限定在此区域。
The apical ectoderm ridges (AERs) are mesodermal stromal cells that induce the formation of protruding structures on their outer ectodermal cells. They are located at the junction of the dorsal and ventral margins of limb buds, and are the major signal centers for the growth and development of limbs. Transgenic technique was used to construct transgenic mice expressing Cre recombinase in AER (Col10a1-Cre). Cre recombinase was specifically expressed in AER so that the target gene anchored by LoxP sequence could be specifically knocked out in AER . The temporal and spatial specificity of the expression of this recombinant mouse Cre recombinase was determined. The experimental results demonstrate that Cre recombinase can be specifically expressed in apical ectoderm ridges and mediate recombination between LoxP. In the apical ectoderm ridges found a specific blue dye, Cre recombinase expression was limited to this area.
