目的研究人乳腺癌细胞中TRIM22对NF-κB表达的影响。方法通过RT-PCR和Western blot方法检测人乳腺癌细胞系MCF-7和人正常乳腺细胞系MCF-10A中TRIM22和NF-κB在mRNA和蛋白水平的表达差异;构建TRIM22表达质粒,并通过瞬时转染TRIM22表达质粒,使MCF-7高表达TRIM22,再检测NF-κB在mRNA及蛋白水平的变化。结果TRIM22在人正常乳腺细胞MCF-10A中的表达水平高于乳腺癌细胞系MCF-7,在MCF-7中转染TRIM22表达质粒后,NF-κB的表达量随之降低。结论在MCF-7细胞中TRIM22可负调控NF-κB的表达。
Objective To investigate the effect of TRIM22 on NF-κB expression in human breast cancer cells. Methods The mRNA and protein levels of TRIM22 and NF-κB in human breast cancer cell line MCF-7 and human normal breast cell line MCF-10A were detected by RT-PCR and Western blot. The TRIM22 expression plasmid was constructed and transiently The TRIM22 expression plasmid was transfected to induce the expression of TRIM22 in MCF-7, and the changes of NF-κB mRNA and protein were detected. Results The expression level of TRIM22 in MCF-10A cells was higher than that in breast cancer cell line MCF-7. The expression of TRIM22 was decreased in MCF-7 cells transfected with TRIM22. Conclusion TRIM22 can negatively regulate the expression of NF-κB in MCF-7 cells.
