目的研究灵芝科假芝属皱盖假芝Amauroderma rude子实体中的化学成分。方法采用正、反相硅胶柱色谱与Sephadex LH-20凝胶柱色谱进行分离纯化,并运用各种波谱方法鉴定化合物结构,采用MTT法测试化合物对人体肿瘤细胞株的细胞毒活性。结果从皱盖假芝子实体95%乙醇提取物中分离鉴定了12个化合物,分别鉴定为diptoindonesin D(1)、6-脱氧巴西红厚壳素(2)、巴西红厚壳素(3)、3-羧基吲哚(4)、3,4-二羟基苯甲酸甲酯(5)、3,4-二羟基苯乙醇(6)、3β-羟基-7,22E-二烯-麦角甾(7)、3β,7α-二羟基-8,22E-二烯-5α,6α-环氧-麦角甾(8)、3β-羟基-7α-甲氧基-8(14),22E-二烯-5α,6α-环氧-麦角甾(9)、5α,8α-过氧麦角固醇(10)、3β,5β,7β-三羟基-6,22E-二烯-麦角甾(11)和3β,5α,6β-三羟基-7,22E-二烯-麦角甾(12)。化合物2与3对人体肿瘤细胞株HL-60、SMMC-7721、A549、MCF-7和SW-480具有一定生长抑制活性,化合物9对HL-60、MCF-7和SW-480具有一定生长抑制活性。结论化合物1~12均为首次从皱盖假芝子实体中分离得到,化合物2、3和9对人体肿瘤细胞株具有一定细胞毒活性。
Objective To study the chemical constituents in the fruiting bodies of Amauroderma rude, Methods The compounds were isolated and purified by normal and reverse phase silica gel column chromatography and Sephadex LH-20 gel column chromatography. The compounds were identified by various spectral methods. The cytotoxic activity of the compounds against human tumor cell lines was tested by MTT assay. Results Twelve compounds were identified from 95% ethanol extract of Rumex japonicus, and were identified as diptoindonesin D (1), 6-deoxy Brazilian red thick chitin (2), Brazilian red thick chitin (3) (4), methyl 3,4-dihydroxybenzoate (5), 3,4-dihydroxyphenethyl alcohol (6), 3β-hydroxy-7,22E-diene- (14), 22E-diene-7α-dihydroxy-8,22E-diene-5α, 6α- (10), 3β, 5β, 7β-trihydroxy-6,22E-diene-ergostat (11) and 3β, 5α, 6β-trihydroxy-7,22E-diene-ergostat (12). Compound 2 and 3 have certain growth inhibitory activity on human tumor cell lines HL-60, SMMC-7721, A549, MCF-7 and SW-480, and compound 9 has certain growth inhibition on HL-60, MCF-7 and SW-480 active. Conclusion Compounds 1 to 12 were isolated from the fruiting body of Rumex niloticus for the first time. Compounds 2, 3 and 9 have some cytotoxic activity on human tumor cell lines.
