目的:研究补肾健脾中药对后巩膜加固术后家兔后巩膜中碱性成纤维细胞生长因子(bFGF)浓度分布的影响,从分子生物学角度研究补肾健脾中药对后巩膜加固术后巩膜组织重塑的影响,进而研究后巩膜加固术结合补肾健脾中药防治高度近视的机理。方法:将成年新西兰白兔12只随机分为空白对照组,单纯手术组(后巩膜加固术组),手术配合中药1组(后巩膜加固配合近视Ⅰ号方组),和手术配合中药2组(后巩膜加固配合近视Ⅱ组)。于术后12周处死部分白兔,摘出右眼球,冲洗干净,切取植片及其附着的巩膜,通过细胞免疫组织化学方法,分析术后白兔后巩膜内bFGF浓度变化。结果:空白组巩膜中bFGF浓度分布非常稀疏,增殖很少;单纯手术组术后初期表现为强烈的炎症反应,有大量炎症细胞浸润,bFGF浓度稀疏,增殖较少;手术配合中药组术后炎症反应较单纯手术组明显较轻,少量炎症细胞浸润,bFGF浓度稠密,大量增殖;手术配合中药1组和手术配合中药2组bFGF浓度分布未见明显差异,均较空白组及单纯手术组密集。结论:后巩膜加固术配合补肾健脾中药能更有效地促进巩膜中bFGF的增殖,从而达到治疗高度近视的作用。
Objective: To study the effect of Bushen Jianpi Chinese medicine on the concentration distribution of bFGF in posterior sclera of rabbits after posterior scleral reinforcement, and to study the effects of Bushen Jianpi Chinese medicine on the distribution of basic fibroblast growth factor (bFGF) Tissue remodeling, and then study the scleral buckling combined with Bushenjianpi Chinese medicine to prevent and control high myopia mechanism. Methods: Twelve adult New Zealand white rabbits were randomly divided into blank control group, simple operation group (posterior scleral reinforcement group), operation with traditional Chinese medicine group 1 (posterior scleral reinforcement plus myopia Ⅰ group), and operation with traditional Chinese medicine group 2 (Posterior scleral reinforcement with myopia Ⅱ group). Some rabbits were sacrificed at 12 weeks after operation. The right eyeballs were excised and rinsed clean. The implants and the sclera attached to them were excised. The bFGF concentration in the sclera of the rabbits after operation was analyzed by immunohistochemistry. Results: The concentration of bFGF in the sclera of the blank group was very sparse and the proliferation was very little. The surgery group had a strong inflammatory reaction at the initial stage, with a large number of infiltration of inflammatory cells, sparse bFGF concentration and less proliferation. The operation combined with the postoperative inflammation Compared with the simple operation group, the response was significantly lighter, a small amount of infiltration of inflammatory cells, bFGF density dense proliferation in large numbers; Conclusion: Posterior Scleral Reinforcement combined with Bushen Jianpi Chinese Medicine can promote the proliferation of bFGF in the sclera effectively, so as to achieve the effect of treating high myopia.
