目的考察由人乳头瘤病毒(human papillomavirus,HPV)主要外壳蛋白(L1)C末端保守序列多肽诱导的单克隆抗体对宫颈脱落细胞内的HPV感染是否具有可应用于临床的检测能力。方法收集三峡大学第二临床医学院妇科门诊宫颈脱落细胞标本,一式两份,其中一份标本用于医院检验科HPV分型检测试剂盒检测,另一份以HPV L1C末端保守序列多肽诱导的小鼠单克隆抗体运用免疫组化方法检测标本中的HPV L1,比较两种方法检测结果的差异。结果收集到的384例妇科门诊宫颈脱落细胞标本中,HPV分型检测试剂盒检测出HPV阳性感染63例,阳性率为16.41%(63/384);用免疫组化检测了其中380例标本,共检出阳性标本120例,阳性检出率为31.58%(120/380),与HPV分型检测试剂盒检测结果相比差异有统计学意义(P<0.05)。结论初步证明了HPV L1C末端保守序列多肽诱导的小鼠单克隆抗体对宫颈脱落细胞内HPV的感染具有良好的检出能力,并且检测能力高于现临床所用的市售HPV分型检测试剂盒。该单克隆抗体在开发用于宫颈癌预防性筛查的HPV检测试剂盒方面具有一定的潜力。
Objective To investigate whether monoclonal antibodies (McAbs) induced by the polypeptide encoded by the conserved C-terminal region of the major coat protein (L1) of human papillomavirus (HPV) have clinical diagnostic capability for HPV infection in cervical exfoliated cells. Methods The samples of cervical exfoliated cells from gynecology clinic of the Second Clinical College of China Three Gorges University were collected in duplicate. One of the specimens was used for the detection of HPV genotyping kit in hospital laboratory and the other one was induced by polypeptide of HPV L1 C-terminal conserved sequence The murine monoclonal antibody was used to detect the HPV L1 in the specimen by immunohistochemistry, and the differences between the two methods were compared. Results Among the 384 cases of exfoliated cervical cytology specimens collected from gynecological clinics, 63 cases were detected positive for HPV infection by HPV typing test kit, with a positive rate of 16.41% (63/384). 380 specimens were detected by immunohistochemistry, A total of 120 positive samples were detected, the positive rate was 31.58% (120/380). The difference was statistically significant (P <0.05) compared with the results of HPV typing test kit. Conclusion The results show that the mouse monoclonal antibody induced by HPV L1 C-terminal conserved sequence peptide has good detection ability for HPV infection in cervical exfoliated cells, and its detection ability is higher than that of commercially available HPV genotyping detection kit. This monoclonal antibody has some potential for developing HPV test kits for the prophylactic screening of cervical cancer.
