目的 探讨人白细胞介素2(IL-2)基因转移对卵巢癌细胞周期和转移生长因子β_1(TGF-β_1)表达的影响。方法 采用流式细胞术探讨IL-2基因转移对卵巢癌细胞SKOV3细胞周期及TGF-β_1表达的影响。结果 SKOV3/IL-2细胞GO、G1期细胞比例明显高于对照组,而S、M期细胞明显少于对照组,P<0.001,表现为G0/G1期阻滞。SKOV3、SKOV3/Neo、SKOV3/IL-2细胞TGF-β_1阳性百分率及荧光强度分别为98.64%、97.00%、99.52%和143、138、163,SKOV3/IL-2平均荧光强度较亲本细胞明显提高,P<0.001,差别非常显著。结论 IL-2基因转移可引起卵巢癌细胞系SKOV3细胞周期G0/G1期阻滞,TGF-β_1表达的增强有可能是IL-2基因转移诱发细胞增殖抑制的原因之一。
Objective To investigate the effect of interleukin 2 (IL-2) gene transfer on the cell cycle and the expression of TGF-β 1 in ovarian cancer cells. Methods Flow cytometry was used to investigate the effect of IL-2 gene transfer on the cell cycle and TGF-β 1 expression in ovarian cancer cell line SKOV3. Results The percentage of cells in GO and G1 phases in SKOV3 / IL-2 cells was significantly higher than that in control cells, while cells in S and M phases were significantly less than those in control cells (P <0.001). The positive percentage of TGF-β 1 and the fluorescence intensity of SKOV3, SKOV3 / Neo and SKOV3 / IL-2 cells were 98.64%, 97.00%, 99.52% and 143,138,163, respectively. The average fluorescence intensity of SKOV3 / IL- , P <0.001, the difference is very significant. Conclusions IL-2 gene transfer may induce cell cycle G0 / G1 arrest in ovarian cancer cell line SKOV3. The enhanced expression of TGF-β1 may be one of the reasons for the inhibition of cell proliferation induced by IL-2 gene transfer.
