目的对IRM-2、ICR、615三种小鼠胸腺(Th)、脾(SP)细胞照射后辐射敏感性的差异进行比较,探讨IRM-2小鼠的辐射抗性及免疫学机制。方法用流式细胞仪检测外周血细胞分型,PA法(FITC-Annexin V和PI标记法)检测Th、SP细胞照射后凋亡率。结果IRM-2小鼠CD4/CD8比值低于ICR、615小鼠;CD25/CD4高于615小鼠(P<0.05),低于ICR小鼠;SP细胞凋亡率0、1、4Gy低于ICR和615小鼠,差异有统计学意义(P<0.05);Th细胞凋亡率低于ICR和615小鼠,在4Gy照射组低于615小鼠(P<0.01)。结论IRM-2小鼠的胸腺和脾细胞对辐射不敏感,辐照后Th、SP细胞凋亡率低于ICR和615小鼠。
Objective To compare the difference of radiosensitivity of thymus (Th) and spleen (SP) cells of IRM-2, ICR, 615 three kinds of mice to investigate the radiation resistance and immunological mechanism of IRM-2 mice. Methods Peripheral blood cell types were detected by flow cytometry. The apoptosis rates of Th and SP cells after irradiation were detected by PA method (FITC-Annexin V and PI-labeled method). Results The CD4 / CD8 ratio of IRM-2 mice was lower than that of ICR 615 mice. CD25 / CD4 was higher than 615 mice (P <0.05), lower than that of ICR mice. The apoptosis rate of SP cells was lower than that of ICR mice ICR and 615 mice, the difference was statistically significant (P <0.05); the apoptosis rate of Th cells was lower than that of ICR and 615 mice, but lower than that of 615 mice in 4Gy irradiation group (P <0.01). Conclusion The thymus and spleen cells of IRM-2 mice are insensitive to radiation. The apoptotic rates of Th and SP cells after irradiation are lower than those of ICR and 615 mice.
