目的:观察降糖消脂片对2型糖尿病(T2DM)模型大鼠的降糖降脂作用,为临床治疗T2DM提供实验依据。方法:采用高脂高糖饲料喂饲大鼠4周,诱发胰岛素抵抗(IR),再按低剂量链脲佐菌素(streptozotocin,STZ)25 mg.kg-1ip,诱发高血糖症,造成T2DM大鼠模型。将实验动物分为正常对照组、模型组、降糖消脂片高、中、低(8,4,2 g.kg-1)剂量组、二甲双胍片(0.2 g.kg-1)组和金芪降糖片(2 g.kg-1)组,连续ig给药8周后杀检,测定空腹血糖(FBG)、总胆固醇(TC)、甘油三酯(TG)、胰岛素(INS)、肿瘤坏死因子(TNF-α)及进行胰腺和肝脏的病理学检查。结果:大鼠在高脂高糖饮食4周时发生高胰岛素血症伴TC增高,成功诱发出IR。STZ ip后造成T2DM大鼠模型。口服8周后,与模型组比较,降糖消脂片高剂量组FBG明显降低(P<0.01),降糖率达到31.23%,INS水平显著增高(P<0.05),TG明显降低(P<0.01),中剂量组TG及TC明显降低(P<0.01~P<0.05),各剂量组TNF-α有降低的趋势;病理结果显示,与模型组比较,高剂量组胰岛数明显增多(P<0.05),中、低剂量组肝脂肪变性明显改善(P<0.05)。结论:降糖消脂片对T2DM模型大鼠有明显的降糖、降脂的作用。
Objective: To observe the hypoglycemic and hypolipidemic effects of Jiangtang Xiaozhi Tablet on type 2 diabetes mellitus (T2DM) rats and provide experimental evidence for clinical treatment of T2DM. Methods: The rats were fed with high-fat and high-sugar diet for 4 weeks to induce insulin resistance (IR). Low-dose streptozotocin (STZ) 25 mg.kg-1 ip was given to induce hyperglycemia and cause T2DM Rat model. The experimental animals were divided into normal control group, model group, hypoglycemic and hypolipidemic high, medium and low dose groups (8,4,2 g.kg-1), metformin tablets (0.2 g.kg-1) The rats in QGD group (2 g · kg-1) were killed after 8 weeks of continuous ig administration. The fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), insulin (INS) Necrosis factor (TNF-α) and pathological examination of the pancreas and liver. Results: Rats with hyperinsulinemia and hypercholesterolemia developed at 4 weeks after high fat and high glucose diet successfully induced IR. STZ ip caused after T2DM rat model. After oral administration for 8 weeks, the FBG of high-dose Jiangtang Xiaozhi Tablet group was significantly lower (P <0.01), the hypoglycemic rate was 31.23%, the level of INS was significantly higher (P <0.05) and the TG significantly lower (P < 0.01). The levels of TG and TC in the middle dose group were significantly decreased (P <0.01 ~ P <0.05), and TNF-α in each dose group decreased. The pathological results showed that compared with the model group, <0.05). Hepatic steatosis was significantly improved in medium and low dose groups (P <0.05). Conclusion: Jiangtang Xiaozhi Tablets has significant hypoglycemic and hypolipidemic effect on T2DM model rats.
