本文是对山东、河南、云南、贵州烟草花叶型病害毒源鉴定工作的一部分。从田间表现畸型、花叶、橡叶症(闪电纹)的植株上获得一种分离物,在心叶烟、蔓陀罗、菜豆(Pinto)上表现局部枯斑。致死温度90℃以上,稀释限点10~(-6),体外保毒期一个月以上。PEG沉淀法获得的提纯物测定紫外分光吸收光谱曲线,符合一般植物病毒核蛋白吸收的特性。电镜观察为刚性长杆状粒体,表明该分离物为典型的TMV。免疫双扩散试验表明:山东、河南、云南、贵州各分离物抗原一致。
This article is part of the identification of tobacco mosaic disease source in Shandong, Henan, Yunnan and Guizhou. An isolate was obtained from plants exhibiting abnormalities in the field, mosaic, and leaf blight (lightning streaks), and showed localized necrosis on leaf tobacco, datura, and Pinto. Lethal temperature above 90 ℃, dilution limit 10 ~ (-6), in vitro protection of more than one month. The purified product obtained by the PEG precipitation method measures the UV-vis absorption spectrum curve, which is in accordance with the absorption characteristic of the general plant virus nucleoprotein. Electron microscopy showed a rigid rod-shaped granular material, indicating that the isolate was a typical TMV. Immune double diffusion test showed that: Shandong, Henan, Yunnan, Guizhou isolates of the same antigen.
