本研究探讨急性白血病(acuteleukemia,AL)细胞株Flt-3基因表达和Flt-3/ITD突变与急性白血病特别是急性髓系白血病(acutemyeloidleukemia,AML)发病的关系。采用RT-PCR分析联合测序的方法检测82例白血病细胞株,其中包括20例AML细胞株和57例急性淋巴细胞白血病细胞株(ALL),5株CML细胞株的Flt-3表达和Flt-3/ITD突变。结果表明:77例AL细胞株中有48例Flt-3表达阳性,阳性表达率为62%,其中20例AML细胞株中有12例阳性,阳性表达率为60%;57例ALL细胞株中有33例阳性,阳性表达率为58%。5例CML细胞株中有3例阳性,阳性表达率为60%。12例Flt-3表达阳性的AML中有1例AMOL细胞株存在有异常表达(阳性率为8.3%),测序显示存在有29bp的两个编码重复序列,其余Flt-3表达阳性的细胞株未检出重复序列。未分化B细胞系Flt-3基因表达阳性率明显高于成熟B细胞ALL(P<0.05)。结论:Flt-3基因在不同种类的白血病细胞中存在着不同程度的表达,在其中1例AML中发现有Flt-3/ITD重复序列。Flt-3基因及Flt-3/ITD突变检测可能有助于ALL特别是AML的诊断。
This study was aimed to investigate the relationship between Flt-3 gene expression and Flt-3 / ITD mutation in acute leukemia (AL) cell line and the pathogenesis of acute leukemia, especially acute myeloid leukemia (AML). 82 leukemia cell lines were detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and sequencing, including 20 AML and 57 acute lymphoblastic leukemia cell lines (ALL), Flt-3 expression and Flt-3 / ITD mutation. The results showed that 48 out of 77 AL cell lines were positive for Flt-3, the positive rate was 62%, of which 12 were positive in 20 AML cell lines and the positive rate was 60%. Among 57 ALL cell lines 33 cases were positive, the positive expression rate was 58%. Three of the five CML cell lines were positive, the positive rate was 60%. In 12 cases of AML with Flt-3 expression, 1 case of AMOL cell line was abnormally expressed (the positive rate was 8.3%). Sequencing showed that there were 29bp two coding repeat sequences, and the remaining Flt-3 positive cell lines did not Duplicate sequences were detected. The positive rate of Flt-3 gene expression in undifferentiated B cell lines was significantly higher than that in mature B cells (P <0.05). Conclusion: Flt-3 gene is expressed in different degree in different types of leukemia cells, Flt-3 / ITD repeats are found in one case of AML. Detection of Flt-3 gene and Flt-3 / ITD mutations may be helpful in the diagnosis of ALL, especially in AML.
