[目的]对影响分裂相指数的各环节进行优化控制,分析该条件下染色体标本制备效果。[方法]在外周血淋巴细胞培养染色体标本制备的基本过程中,对细胞培养、细胞周期阻滞、细胞收获等步骤中与分裂相指数相关的各环节进行优化控制,采用显微镜下直接观察、计数及染色体分散面积测量等方法对控制后的标本制备效果进行分析。[结果]在优化条件下所得染色体标本分裂相数较多、染色体分散良好、制备效果稳定。[结论]该优化控制方案可获得理想的可供分析的染色体分裂相,效果稳定,保证了外周血淋巴细胞染色体制备及分析的成功率。
[Objective] The optimization and control of the links affecting the fission phase index were carried out to analyze the preparation effect of chromosome specimens. [Methods] During the process of preparation of chromosomal samples of peripheral blood lymphocytes, all steps related to mitotic index in cell culture, cell cycle arrest, cell harvesting and other steps were optimized and controlled. The cells were directly observed under the microscope and counted And chromosomal scatter area measurement and other methods to control the preparation of specimens for analysis. [Result] The chromosomes obtained under the optimal conditions had more chromosomes, better chromosomes and better stability. [Conclusion] The optimal control scheme could obtain ideal chromosomal splitting phase for analysis, the effect was stable, and the success rate of chromosome preparation and analysis of peripheral blood lymphocytes was guaranteed.
