胚胎与子宫内膜上皮细胞之间的黏附是胚胎成功植入的关键.岩藻糖基转移酶Ⅳ(FUT4)对胚胎细胞与子宫内膜细胞黏附的影响未见报道.本研究以人子宫内膜细胞(HEC-1A)和胚胎细胞(JAR)为体外着床模型,观察上调HEC-1A细胞中FUT4表达对JAR细胞与HEC-1A细胞黏附的影响.RT-PCR和免疫细胞化学检测结果显示,FUT4过表达增加HEC-1A细胞中FUT4基因及蛋白的表达;免疫细胞化学及Western印迹结果表明,上调HEC-1A细胞中FUT4增加细胞表面LeY的合成;细胞黏附实验结果显示,与未转染组相比较,FUT4过表达增加了JAR细胞与HEC-1A细胞的黏附率.本研究证明,FUT4过表达可以增加细胞表面LeY寡糖抗原的合成,从而促进胚胎细胞与子宫内膜细胞的黏附.
The adhesion between embryo and endometrial epithelial cells is the key to the successful implantation of embryos.Fucosyltransferase Ⅳ (FUT4) on the adhesion of embryonic cells and endometrial cells has not been reported.In this study, human uterus The effects of FUT4 expression in HEC-1A cells on the adhesion of JAR cells to HEC-1A cells were observed by using HEC-1A and JAR as in vitro models.RT-PCR and immunocytochemistry , FUT4 overexpression increased the expression of FUT4 gene and protein in HEC-1A cells. Immunocytochemistry and Western blotting showed that up-regulation of FUT4 increased the cell surface LeY synthesis in HEC-1A cells. The results of cell adhesion assay showed that, compared with untransfected Overexpression of FUT4 increased the adhesion rate of JAR cells to HEC-1A cells.This study demonstrated that FUT4 overexpression can increase the synthesis of cell-surface LeY oligosaccharide antigen and promote the adhesion of embryonic cells to endometrial cells.
