目的 :建立测定人血清中酒石酸美托洛尔浓度的HPLC方法 ,为临床药理研究提供科学的分析技术。方法 :血清样品经萃取纯化后溶于流动相中 ,进样分析。测定使用C18色谱柱 ,甲醇 水相 ( 50∶50 ,水相中含 0 .1 6%戊烷磺酸钠和 0 .0 1 4 %无水乙酸钠 )为流动相 ,以纳多洛尔为内标 ,用荧光检测Ex =2 77nm ,Em =2 99nm。结果 :本方法线性范围为 1 0~ 30 0ng/ml,最低检测浓度为 1ng/ml。平均回收率为 ( 1 0 2 .1 5± 6.76) %。结论 :本方法选择性强 ,灵敏 ,重现性好 ,适用于临床血药浓度检测
OBJECTIVE: To establish a HPLC method for the determination of metoprolol tartrate in human serum and to provide a scientific analytical technique for clinical pharmacology research. Methods: The serum samples were purified by extraction and dissolved in the mobile phase, and injected for analysis. The mobile phase consisted of C18 column, aqueous methanol (50:50, 0.1% sodium pentane sulfonate and 0.014% sodium acetate anhydrous) as the mobile phase and nadolol as Internal standard, with fluorescence detection Ex = 2 77nm, Em = 299nm. Results: The linear range of this method was 10-30ng / ml and the lowest detection concentration was 1ng / ml. The average recovery was (102.1 ± 6.76)%. Conclusion: The method is selective, sensitive and reproducible, and is suitable for clinical blood concentration detection
